I suppose you need some Gtk+ bases. About C++, its use is very basic: classes, inheritance (simple and multiple). The best way is probably to read some code starting from plugins which implement the various tools.
Gtkmm and Qt are not options, at least for now.

Thank you for your answer. I am not a programmer but I start to learn c++ now and maybe in future I will help you in coding gchempaint. I know you don't use gtkmm and qt in gchempaint...
What do I should learn in c++ to understand and write gchempaint source code? Is it possible to use gtkmm or qt?
Excuse me if I said something wrong. I am from Russia and I am learning English too.

My english is not better than yours.
I suppose it can work for mac OS-X, things will be probably more involved for Windows. If a volunteer wants to try I'll help him as far as I can, but I am not an expert of theses systems.
